Design Notes

ARK-shRNA-1 template sequence starts with GG, which is for transcription efficiency. GC content is around 50%, its target site is not in the first 75 bases after AUG, these are for the RNAi efficiency. UU overhangs in 3′-end, this is for the stability of shRNA. The antisense strand of shRNA is reverse complement with the sequence of the target site.

ARK-shRNA-1 template is designed from the sequence of the target site in Arginine Kinase mRNA of Phyllotreta striolata, which is 997-1018 after AUG. shRNA contains a sense strand, a loop, and an antisense strand.
